How'd you know it's possible? I can see some colored squares in screenshot that might be it, but

Apparently you can unlock them by using only certain functions--seems like a lot of boring
waiting around to me, though. Copy-pasting directions to efficient unlocking:
1: Rainbow Dash - Pump: Off Vibrator: Fast - (Unlocks Pinkie)
2: Pinkie Pie - Pump: Fast Vibrator: Off - (Unlocks Applejack, Derpy)
3: Applejack - Pump: Medium Vibrator: Slow - (Unlocks Fluttershy)
4: Pinkie Pie - Pump: Medium Vibrator: Fast - (Unlocks Rarity)
5: Rarity - Pump: Fast Vibrator: Fast - (Unlocks None)
6: Fluttershy - Pump: Fast Vibrator: Fast - (Unlocks Twilight)

Unlocking All
It's pretty damned easy to unlock all of them.
After decompiling the swf, it's pretty straightforward.
Pinkie Pie: Only use vibration
Rarity: Overflow the "milk" collector
Applejack: Only use the pump
Fluttershy: Don't use the highest settings
Derpy Hooves: Orgasm, but don't cum
Twilight Sparkle: Orgasm with all other characters
Multiple Orgasms: Orgasm and fill the container with all other characters

How do you fill the container with each character? The only one I've managed with so far is

easier said than done.

All you have to do is get the orgasm meter almost to the very top, then stop. It'll shot the cum
meter way up. Just keep doing that until you fill the collector.
