He knows Jack's quest is hopeless, aside from possibly killing Aku in the future. And even that is unlikely. And he just wants to convince Jack, that maybe they don't have to kill each other because ultimately, Aku just wants control, not genocide. And Mako sells it. That's the key. He sells it and he sells it damn well. For all of 5 seconds, Jack has his moment of doubt. And in a children's cartoon, 5 seconds of genuine doubt is a goddamn eternity.
